  • Endonuclease Q (EndoQ) is a recently discovered enzyme from the hyperthermophilic archaeon Pyrococcus furiosus by Ishino's group. EndoQ can specifically recognize and cut the DNA backbone at the 5'-side of deaminated bases (uracil, hypoxanthine, xnathine) and abasic site. EndoQ was also found to interact with PCNA, a sliding clamp, in P. furiosus, and thus, EndoQ has been considered as a DNA repair enzyme. However, the repair pathway madiated by EndoQ is still little understood; e.g. which polymerase is involved, how to remove the lesion. In this study, we address the fully understanding of the EndoQ-mediated DNA repair pathway by identifying the interacting partner of EndoQ from P. furiosus.
